چکیده

With their potent regenerative and protective capacities, stem cell-derived conditioned media emerged as an effective alternative to cell therapy, have a prospect be manufactured pharmaceutical products for tissue regeneration applications. Our study investigates the neuroregenerative potential of human dental pulp cells (DPCs) medium (CM) defines optimization strategy DPC-CM enhanced neuronal outgrowth. Primary sensory neurons from mouse dorsal root ganglia were cultured with or without DPC-CM, lengths ?III-tubulin positive neurites measured. The impacts several manufacturing features duration conditioning, CM storage, preconditioning DPCs some factors on functional activity assessed neurite length. We observed that significantly outgrowth in concentration-dependent manner. frozen storage had no impact experimental outcomes 48 h DPC conditioning is optimal CM. To further understand feature we studied secretome by growth factor antibody array analysis revealed presence involved either neurogenesis, neuroprotection, angiogenesis, osteogenesis. B-27 supplement effect changing its composition factors. Here, show stimulate primary neurons. Moreover, identified secreted protein candidates can potentially promote this promising DPC-CM.

Dental pulp-derived stem cell conditioned medium reduces cardiac injury following ischemia-reperfusion

Stem cells from human exfoliated deciduous teeth (SHEDs) can regenerate various tissues. We investigated the impact of SHED-conditioned medium (SHED-CM) on myocardial injury in a mouse model of ischemia-reperfusion (I/R). Wild-type (WT) mice were subjected to myocardial ischemia followed by reperfusion. SHED-CM was intravenously injected at 5 min after reperfusion. Administration of SHED-CM red...

Differentiation of dental pulp stem cells into regular-shaped dentin-pulp complex induced by tooth germ cell conditioned medium.

Investigations of the odontoblast phenotype are hindered by obstacles such as the limited number of odontoblasts within the dental pulp and the difficulty in purification of these cells. Therefore, it is necessary to develop a cell culture system in which the local environment is inductive and can promote dental pulp stem cells (DPSCs) to differentiate into odontoblast lineage. In this study, w...

Conditioned medium from human bone marrow stromal cells attenuates initial inflammatory reactions in dental pulp tissue.

AIM To evaluate the effect of MSC-conditioned medium (CM) on the secretion of pro- and anti-inflammatory cytokines from dental pulp cells (hDPC) in vitro, and on the gene expression in vivo after replantation of rat molars. MATERIALS AND METHODS hDPC were cultured in CM for 24 h, and the concentration of interleukin IL-10, IL-4, IL-6, and IL-8, regulated on activation, normal T Cell expressed...

The Effects of Dental Pulp Stem Cell Conditioned Media on the Proliferation of Peripheral Blood Mononuclear Cells

Background: Dental Pulp Stem Cells (DPSCs) are multipotent mesenchymal stem cells. DPSCs can renew themselves and differentiate into various cell types such as adipocytes, osteocytes, neurons, etc. DPSCs possess immunomodulatory properties and can inhibit peripheral blood mononuclear cell (PBMC) proliferation. Recent studies showed that conditioned-medium mesenchymal stem cells also had immunos...
